Background Pink bollworm,Pectinophora gossypiella(Saunders)(Lepidoptera:Gelechiidae)has become a poten-tial pest of cotton by causing substantial yield losses around the world including Pakistan.Keeping in view the fa...Background Pink bollworm,Pectinophora gossypiella(Saunders)(Lepidoptera:Gelechiidae)has become a poten-tial pest of cotton by causing substantial yield losses around the world including Pakistan.Keeping in view the facts like limited research investigations,unavailability,and high cost of artificial diet’s constituents and their premixes,the present research investigations on the dietary aspect of P.gossypiella were conducted.The larvae of P.gossypiella were reared on different diets that were prepared using indigenous elements.The standard/laboratory diet com-prised of wheat germ meal 34.5 g,casein 30.0 g,agar–agar 20.0 g,sucrose 10.0 g,brewer’s yeast 5.0 g,α-cellulose 1.0 g,potassium-sorbate1.5 g,niplagin 0.5 g,decavitamin 0.01 g,choline-chloride 0.06 g,maize-oil 3.30 g,honey 2.0 g,and water 730.0 mL.Alternatives to cotton bolls and wheat germ meal were okra seed sprouts,okra fruit,cottonseed meal,and okra seed meals,which were included in the study to introduce an efficient and economic mass-rearing system.Results The larval development completed in 19.68d±0.05 d with a weight of 20.18mg±0.20 mg at the fourth instar fed on the cottonseed meal-based diet instead of wheat germ meal based diet.On the same diet,84.00%±4.00%,17.24 mg±0.03 mg,and 7.76d±0.06 d were recorded as pupae formation,pupal weight,and pupal duration,respectively.Adult emergence,76.00%±1.00%was recorded from pupae collected from larvae raised on cottonseed meal-based diet.These male and female moths lived for 40.25d±0.10 d,and 44.34d±0.11 d,respectively.Females deposited 21.28±0.04 eggs per day with the viability of 65.78%±0.14%.The larval mortal-ity at the fourth instar was 37.20%±1.36%and malformed pupation of 12.00%±1.41%was recorded.Replacement of wheat germ meal with that of local meals(cottonseed and okra seed)in the standard laboratory diet has saved 463.80 to 467.10 PKR with 1.62 to 1.63 cost economic returns,respectively.Conclusion This research is of novel nature as it provides a concise and workable system for the economic and suc-cessful rearing of P.gossypiella under laboratory conditions.展开更多
Background SRO(Similar to RCD1)genes family is largely recognized for their importance in the growth,develop-ment,and in responding to environmental stresses.However,genome-wide identification and functional character...Background SRO(Similar to RCD1)genes family is largely recognized for their importance in the growth,develop-ment,and in responding to environmental stresses.However,genome-wide identification and functional characteri-zation of SRO genes from cotton species have not been reported so far.Results A total of 36 SRO genes were identified from four cotton species.Phylogenetic analysis divided these genes into three groups with distinct structure.Syntenic and chromosomal distribution analysis indicated uneven distribu-tion of GaSRO,GrSRO,GhSRO,and GbSRO genes on A2,D5 genomes,Gh-At,Gh-Dt,Gb-At,and Gb-Dt subgenomes,respectively.Gene duplication analysis revealed the presence of six duplicated gene pairs among GhSRO genes.In promoter analysis,several elements responsive to the growth,development and hormones were found in GhSRO genes,implying gene induction during cotton growth and development.Several miRNAs responsive to plant growth and abiotic stress were predicted to target 12 GhSRO genes.Organ-specific expression profiling demonstrated the roles of GhSRO genes in one or more tissues.In addition,specific expression pattern of some GhSRO genes dur-ing ovule development depicted their involvement in these developmental processes.Conclusion The data presented in this report laid a foundation for understanding the classification and functions of SRO genes in cotton.展开更多
Background Cotton is a strategically important fibre crop for global textile industry.It profoundly impacts several countries’industrial and agricultural sectors.Sustainable cotton production is continuously threaten...Background Cotton is a strategically important fibre crop for global textile industry.It profoundly impacts several countries’industrial and agricultural sectors.Sustainable cotton production is continuously threatened by the unpre-dictable changes in climate,specifically high temperatures.Breeding heat-tolerant,high-yielding cotton cultivars with wide adaptability to be grown in the regions with rising temperatures is one of the primary objectives of modern cotton breeding programmes.Therefore,the main objective of the current study is to figure out the effective breed-ing approach to imparting heat tolerance as well as the judicious utilization of commercially significant and stress-tolerant attributes in cotton breeding.Initially,the two most notable heat-susceptible(FH-115 and NIAB Kiran)and tolerant(IUB-13 and GH-Mubarak)cotton cultivars were spotted to develop filial and backcross populations to accom-plish the preceding study objectives.The heat tolerant cultivars were screened on the basis of various morphological(seed cotton yield per plant,ginning turnout percentage),physiological(pollen viability,cell membrane thermostabil-ity)and biochemical(peroxidase activity,proline content,hydrogen peroxide content)parameters.Results The results clearly exhibited that heat stress consequently had a detrimental impact on every studied plant trait,as revealed by the ability of crossing and their backcross populations to tolerate high temperatures.However,when considering overall yield,biochemical,and physiological traits,the IUB-13×FH-115 cross went over particularly well at both normal and high temperature conditions.Moreover,overall seed cotton yield per plant exhibited a posi-tive correlation with both pollen viability and antioxidant levels(POD activity and proline content).Conclusions Selection from segregation population and criteria involving pollen viability and antioxidant levels concluded to be an effective strategy for the screening of heat-tolerant cotton germplasms.Therefore,understanding acquired from this study can assist breeders identifying traits that should be prioritized in order to develop climate resilient cotton cultivars.展开更多

Using the UALCAN informational index, GINS1 expression assessment unveiled a critical up- regulation in malignant cells that stood out from normal controls, suggesting its contribution to COAD expansion. Further dismantling GINS1 expression across various boundaries revealed unsurprising up-regulation in different malignant development stages, racial groups, genders, and age classes in COAD patients, characteristics for its imperative role in cancer progression. Moreover, this study investigated the promoter methylation status of GINS1, uncovering a critical uniqueness between COAD samples and normal controls. Analyzing promoter methylation across various clinical boundaries uncovered powerful variations, with particular methylation patterns seen across cancer stages, race groups, genders, and age groups. Survival analysis using the Kaplan-Meier (KM) plotter tool showed a colossal connection between GINS1 expression levels and overall survival (OS) in COAD patients, with low GINS1 expression interfacing with higher OS. Additionally, mutational examination using the cBioPortal stage revealed that no critical change was found in COAD. Overall, these findings revealed the complex contribution of GINS1 in COAD pathogenesis, underlining its actual limit as a prognostic biomarker and supportive therapeutic agent in COAD management.展开更多

A field study was carried out to search out the suitable rice transplanting time for four different coarse genotypes under the semi-arid environment of Faisalabad. The experiment was conducted at Agronomic Research Area, University of Agriculture, Faisalabad and was laid out in randomized complete block design (RCBD) with split plot arrangement keeping transplanting time in main plots while rice genotypes in subplots. Variability among treatments was measured by Fisher’s ANOVA (P ≤ 5%) and LSD test was applied to compare the differences among treatments’ means. The ANOVA indicated statistically significant differences among genotypes as well as transplanting dates irrespective of all studied traits while interactive effects of both were found to be non-significant. NIBGE-1 performed best with maximum paddy yield of 6.05 t/ha while KSK-434 performed poor with paddy yield of 2.78 t/ha. Increased paddy yield and yield related parameters of all rice genotypes were recorded where transplantation was done on 25th of June. Generally, paddy yield decreased with delaying the transplanting time. The results suggested that NIBGE-1 can perform better under the semi-arid conditions of Faisalabad and last week of June might be the optimum time for nursery transplantation. It can also be further elucidated that late transplanting causes yield reduction which could not be recommended among farmers.展开更多

Besides other pollutants, high concentration of uranium above a certain level in drinking water is also hazardous. As water and food are the main sources of uranium intake, it is absolutely imperative that their level of concentrations is monitored regularly for safety of the general public. In this regard, trace amount of uranium in drinking water samples collected from different locations of Faisalabadcity has been carried out using Neutron Induced Fission Track Technique. The water samples along with the standard of known uranium concentration were poured and dried over Lexan track detectors and then irradiated with thermal neutrons in Reactor. After etching, the tracks produced in the detectors as a result of 235U (n, f) reaction were counted under an optical microscope. The uranium concentration was determined by counting and comparing the observed fission track density in the samples and the standard. The observed uranium concentration in the studied water samples varied from (1.04 ± 0.30) μg·L-1 to (21.08 ± 2. 95) μg·L-1 with an average value of (7.39 ± 1.30) μg·L-1 and is found to be within safe limits as far as uranium related health hazards are concerned.展开更多
Background High temperature stress at peak flowering stage of cotton is a major hindrance for crop potential.This study aimed to increase genetic divergence regarding heat tolerance in newly developed cultivars and hy...Background High temperature stress at peak flowering stage of cotton is a major hindrance for crop potential.This study aimed to increase genetic divergence regarding heat tolerance in newly developed cultivars and hybrids.Fifty cotton genotypes and 40 F1(hybrids)were tested under field conditions following the treatments,viz.,high temperature stress and control at peak flowering stage in August and October under April and June sowing,respectively.Results The mean squares revealed significant differences among genotypes,treatments,genotype×treatment for relative cell injury,chlorophyll contents,canopy temperature,boll retention and seed cotton yield per plant.The genetic diversity among 50 genotypes was analyzed through cluster analysis and heat susceptibility index(HSI).The heat tolerant genotypes including FH-Noor,NIAB-545,FH-466,FH-Lalazar,FH-458,NIAB-878,IR-NIBGE-8,Weal-AGShahkar,and heat sensitive,i.e.,CIM-602,Silky-3,FH-326,SLH-12 and FH-442 were hybridized in line×tester fashion to produce F1 populations.The breeding materials’populations(40 F1)revealed higher specific combining ability variances along with dominance variances,decided the non-additive type gene action for all the traits.The best general combining ability effects for most of the traits were displayed by the lines,i.e.,FH-Lalazar,NIAB-878 along with testers FH-326 and Silky-3.Specific combining ability effects and better-parent heterosis were showed by the crosses,viz.,FH-Lalazar×Silky-3,FH-Lalazar×FH-326,NIAB-878×Silky-3,and NIAB-878×FH-326 for seed cotton yield and yield contributing traits under high temperature stress.Conclusion Heterosis breeding should be carried out in the presence of non-additive type gene action for all the studied traits.The best combiner parents with better-parent heterosis may be used in crossing program to develop high yielding cultivars,and hybrids for high temperature stress tolerance.展开更多
Background Under K deficiency the uptake and distribution pattern in plant cells is mediated through different transport proteins and channels which were controlled by specific gene family.Therefore,a hydroponic exper...Background Under K deficiency the uptake and distribution pattern in plant cells is mediated through different transport proteins and channels which were controlled by specific gene family.Therefore,a hydroponic experiment was conducted under control condition for testing the gene expression pattern of the K transporter under adequate and low K supply levels.After that,a 2-year field experiment was conducted to evaluate five selected cotton cultivars(four K-efficient cultivars,viz.,CIM-554,CYTO-124,FH-142,IUB-2013,and one K non-efficient,BH-212) screened from the initial hydroponics culture experiment and two levels of potassium(0 K_(2)O kg·ha^(-1) and 50 K_(2)O kg·ha^(-1)) were tested under reduced irrigation(50% available water content;50 AWC) and normal irrigation conditions(100% available water content;100 AWC).Result Results revealed that the transcript levels of GhHAK5aD in roots were significantly higher in K^(+) efficient cultivars than that in K^(+) non-efficient cultivars.The GhHAK5aD expression upon K^(+) deficiency was higher in roots but lower in shoots,indicating that GhHAK5aD could have a role in K^(+) uptake in roots,instead of transport of K^(+) from root to shoot.Similarly,under field conditions the cultivar FH-142 showed an increase of 22.3%,4.9%,2.4%,and 1.4% as compared with BH-212,IUB-2013,CYTO-124,and CIM-554,respectively,in seed cotton yield(SCY) with K application under reduced irrigation conditions.With applied K,the FH-142 showed an increase in net photosynthetic rate by 57.3% as compared with the rest of the cultivars under reduced irrigation over K control.However,the overall performance indicators of K-efficient cultivars like FH-142,CYTO-124,CIM-554,and IUB-2013 were better than BH-212(K in-efficient) under reduced irrigation conditions with applied K at 50 kg·ha^(-1).Fiber quality trait improved significantly with K application under water deficit.The increase in micronaire was 3.6%,4.7%,7.8%,3.4%,and 6.7% in BH-212,IUB-2013,CIM-554,CYTO-124,and FH-142,respectively,with K application at 50 kg·ha^(-1) over without K application under reduced irrigation conditions during the cotton growing season.Similarly,the cultivars FH-142 increased by 12% with K application under reduced irrigation as compared with other cultivars.The performance of K-efficient cultivars under reduced irrigation conditions was 30% better in SCY and quality traits with the application of K at 50 kg·ha^(-1) as compared with K-non-efficient cultivars.Similarly,water use efficiency(WUE)(40.1%) and potassium use efficiency(KUE)(20.2%) were also noted higher in case of FH-142 as compared with other cultivar with K application under reduced conditions.Conclusion Higher expression of GhHAK5aD gene was observed in K-efficient cultivars as compared with K-nonefficient cultivars in roots indicates that GhHAK5aD may be contributing to genotypic differences for K^(+) efficiency in cotton.K-efficient cotton cultivars can be used for the low-K environments and can also be recommended for general cultivars.展开更多

Fruitfully, lemon juice which contains various acidic fun...In this research study, we have synthesized the bio-capped ZnO/g-C_(3)N_(4) nanocomposites by employing lemon juice(Citrus limon) as a stabilizer and mediator. Fruitfully, lemon juice which contains various acidic functional groups and citric acid has the capability to block the surface of g-C_(3)N_(4) from chemical reactivity and activated the surface of g-C_(3)N_(4) for various reactions. Consequently, the agglomeration behavior and controlled shape of g-C_(3)N_(4) has also been achieved. Our experimental results i.e. XRD,TEM, HRTEM, PL, FS, XPS, and PEC have confirmed that the lemon juice mediated and green g-C_(3)N_(4)(L-CN) have good performances and remarkable visible light photocatalytic activities as compared to the chemically synthesized g-C_(3)N_(4)(CN). Furthermore, the small surface area and low charge separation of g-C_(3)N_(4) is upgraded by coupling with Zn O nanoparticles. It is proved that the coupling of Zn O worked as a facilitator and photoelectron modulator to enhance the charge separation of g-C_(3)N_(4). Compared to pristine lemon-mediated green g-C_(3)N_(4)(L-CN), the most active sample 5Zn O/L-CN showed ~ 5-fold improvement in activities for ciprofloxacin(CIP) and methylene blue(MB) degradation. More specifically,the mineralization process and degradation pathways, and the mineralization process of ciprofloxacin(CIP) and methylene blue(MB) are suggested. Finally, our present novel research work will provide new access to synthesize the eco-friendly and bio-caped green g-C_(3)N_(4)nanomaterials and their employment for pollutants degradation and environmental purification.展开更多

Nitrogen and water are two most important yield limiting factors which effect economic production of wheat. Drought initiates when large area of land suffers from absence of precipitation for temporary periods. Though, not only water scarceness but also low relative humidity and high temperature are other climate factors which induce drought condition. Whereas Effective nutrition (nitrogen) maintains crops metabolic activities and has the potential to lessen drought stress. According to study carried out to explore the influence of varying levels of potential soil moisture deficit (PSMD) and nitrogen, an experiment was planned at agronomy research area, University of Agriculture, Faisalabad. It was laid out in randomized complete block design (RCBD) with split plot arrangement, and replicated thrice. Two factors: potential soil moisture deficit levels (I1: irrigation at 50 mm PSMD, I2: irrigation at 75 mm PSMD and I3: irrigation at 100 mm PSMD) were randomized in main plots, while sub plot nitrogen levels {control (No nitrogen), 50, 100 and 150 kg N ha-1} were randomized. Adequate nitrogen application along with sufficient irrigation management gave highest yield. Under water deficit conditions adequate nitrogen application ameliorates the drought stress. Highest plant height (86.27 cm), number of productive tillers (320.0), grain·spike-1(49.73), test weight (50.55 g) and grain yield (6.72 t·ha-1) were observed with 75 mm PSMD and 150 kg·ha-1 application of nitrogen. While radiation use efficiency for dry matter accumulation of wheat ranged from 1.53 - 1.87 g·MJ-1 whereas, radiation use efficiency for grain yield ranged from 0.421 - 0.473 g·MJ-1. Results revealed that highest RUE obtained I3 (75 mm PSMD) and nitrogen application at 150 kg·ha-1. Judicial application of irrigation and fertilizer not only boosts the yield but also saves resource and increases former output.展开更多
Background Boron(B)deficiency is an important factor for poor seed cotton yield and fiber quality.However,it is often missing in the plant nutrition program,particularly in developing countries.The current study inves...Background Boron(B)deficiency is an important factor for poor seed cotton yield and fiber quality.However,it is often missing in the plant nutrition program,particularly in developing countries.The current study investigated B’s effect on growth,yield,and fiber quality of Bt(CIM-663)and non-Bt(Cyto-124)cotton cultivars.The experimental plan consisted of twelve treatments:Control(CK);B at 1 mg·kg^(−1) soil application(SB1);2 mg·kg^(−1) B(SB2);3 mg·kg^(−1) B(SB3);0.2%B foliar spray(FB1);0.4%B foliar spray(FB2);1 mg·kg^(−1) B+0.2%B foliar spray(SB1+FB1);1 mg·kg^(−1) B+0.4%B foliar spray(SB1+FB2);2 mg·kg^(−1) B+0.2%B foliar spray(SB2+FB1);2 mg·kg^(−1) B+0.4%B foliar spray(SB2+FB2);3 mg·kg^(−1) B+0.2%B foliar spray(SB3+FB1);3 mg·kg^(−1) B+0.4%B foliar spray(SB3+FB2).Each treat-ment has three replications,one pot having two plants per replication.Results B nutrition at all levels and methods of application significantly(P≤0.05)affected the growth,physiological,yield,and fiber quality characteristics of both cotton cultivars.However,SB2 either alone or in combination with foliar spray showed superiority over others,particularly in the non-Bt cultivar which responded better to B nutrition.Maxi-mum improvement in monopodial branches(345%),sympodial branches(143%),chlorophyll-a(177%),chlorophyll-b(194%),photosynthesis(169%),and ginning out turn(579%)in the non-Bt cultivar was found with SB2 compared with CK.In Bt cultivar,although no consistent trend was found but integrated use of SB3 with foliar spray performed relatively better for improving cotton growth compared with other treatments.Fiber quality characteristics in both cultivars were improved markedly but variably with different B treatments.Conclusion B nutrition with SB2 either alone or in combination with foliar spray was found optimum for improving cotton’s growth and yield characteristics.展开更多
Salinity is one of the major abiotic factors that limit the growth and productivity of plants.Foliar application of plant growth regulators(PGRs)may help plants ameliorate the negative impacts of salinity.Thus,a field...Salinity is one of the major abiotic factors that limit the growth and productivity of plants.Foliar application of plant growth regulators(PGRs)may help plants ameliorate the negative impacts of salinity.Thus,a field experiment was conducted at the Botanical Garden University of Balochistan,Quetta,to explore the potential role of PGRs,i.e.,moringa leaf extract(MLE;10%),proline(PRO;1μM),salicylic acid(SA;250μM),and thiourea(TU;10 mM)in ameliorating the impacts of salinity(120 mM)on Plantago ovata,an important medicinal plant.Salinity hampered plant photosynthetic pigments and metabolites but elevated oxidative parameters.However,foliar application of PGRs enhanced photosynthetic pigments,including Chl b(21.11%),carotenoids(57.87%)except Chl a,activated the defense mechanisms by restoring and enhancing the metabolites,i.e.,soluble sugars(49.68%),soluble phenolics(33.34%),and proline(31.47%),significantly under salinity stress.Furthermore,foliar supplementation of PGRs under salt stress led to a decrease of about 43.02%and 43.27%in hydrogen peroxide and malondialdehyde content,respectively.Thus,PGRs can be recommended for improved photosynthetic efficiency and metabolite content that can help to get better yield under salt stress,with the best and most effective treatments being those of PRO and MLE to predominately ameliorate the harsh impacts of salinity.展开更多
We developed a molecular marker for MAS of mungbean resistant varieties against CLS from the consensus sequence(MB-CLsRG)of identified RGAs(MB-ClsRCaG1 and MB-ClsRCaG2).The MB-CLsRG sequence-specific primer pair was u...We developed a molecular marker for MAS of mungbean resistant varieties against CLS from the consensus sequence(MB-CLsRG)of identified RGAs(MB-ClsRCaG1 and MB-ClsRCaG2).The MB-CLsRG sequence-specific primer pair was used to screen Cercospora leaf spot(CLS)resistant varieties of mungbean in genomic analysis that showed congruency with phenotypic screening.Validation of molecular marker linkage with CLS resistance was performed using rtPCR in transcriptomic analysis.The sequenced PCR products showed 100%homology with MB-CLsRG sequence and putative disease resistance proteins that confirmed the linkage of molecular marker with CLS resistance in mungbean.The antifungal potential of MB-CLsRG gene encoding protein was assessed.The MB-CLsRG gene sequence was cloned in the E.coli expression vector for recombinant protein production.The recombinant protein was then investigated for its in vitro antifungal potential against Cercospora canescens.The in vitro investigation showed strong antifungal activity of recombinant protein as it restricted the growth of fungal mycelial mass.The results validated the linkage of developed marker with CLS-resistant mungbean varieties;therefore,it can be used to screen resistant varieties from a large population in MAS.Moreover,the recombinant protein of the MB-CLsRG gene sequence revealed antifungal potential,which proved the gene sequence could be suitable to use in transgenic plants technology to develop fungal-resistant transgenic crops.展开更多
Calotropis procera(Aiton)W.T.Aiton,belonging to the family Apocynaceae,is C3 evergreen plant species in arid and semi-arid areas of the Punjab Province,Pakistan.It grows in a variety of habitats like salt affected and...Calotropis procera(Aiton)W.T.Aiton,belonging to the family Apocynaceae,is C3 evergreen plant species in arid and semi-arid areas of the Punjab Province,Pakistan.It grows in a variety of habitats like salt affected and waterlogged area,desert/semi-desert,roadside,wasteland,graveyard,forest,crop field,coastline,and river/canal bank.A total of 12 populations growing in different ecological regions were sampled to evaluate their growth,physio-biochemical,and anatomical responses to specific environmental condition.Population adapted to desert/semi-desert showed vigorous growth(plant height,shoot length,and number of leaves),enhanced photosynthetic level(chlorophyll a,chlorophyll b,carotenoids,and total chlorophyll),and apparent anatomical modifications such as increased stem radius,cuticle thickness,storage parenchyma tissues(cortex and pith),and vascular bundles in stems,while the maximum of midrib and lamina thickness,epidermal cells,cuticle thickness,cortical proportion,abaxial stomatal density,and its area in leaves.There was high plasticity in structural and functional features of these populations,which enable them to survive and tolerate under such hot and dry desert environment.Population of saline areas exhibited very critical modifications to sustain under salt prone environment.At physiological level,it possesses the maximum amount of organic osmolytes(glycine betaine and proline)and antioxidants(superoxide dismutase(SOD),catalase(CAT),and peroxidase(POD)),while at anatomical level,it showed intensive sclerification,large phloem region(inner and outer),pith parenchyma cells,and metaxylem vessels in stems and leaves.The population of dry mountains showed very distinctive features,such as increased shoot ionic contents(K+and Ca2+),collenchyma and sclerenchyma thickness in stems,trichomes size,and numerous small stomata on abaxial surface of leaves.It is concluded that no definite or precise single character can be taken as a yardstick for adjudging the biomass production in this rubber bush weed population.展开更多

Chrysopogon serrulatus(false beard-grass)is a dominant component of vegetation in the foothills of the Himalayas.To study whole plant morphology,individuals of C.serrulatus were collected from three plots at each of s...Chrysopogon serrulatus(false beard-grass)is a dominant component of vegetation in the foothills of the Himalayas.To study whole plant morphology,individuals of C.serrulatus were collected from three plots at each of six locations spanning from 400 to 1,400 m.The population colonizing the highest elevation modifications in different plant organs.Roots showed increased metaxylem number and area.In the stem,especially outside of the vascular tissue,there was intensive sclerification indicative of increased xeromorphy as a survival strategy.At the highest elevation,leaves were wider;aerenchyma formation and increased sclerification were noted in the leaf sheath;and a greater proportion of storage parenchyma was observed in the leaf blade,all indicators of succulence.In contrast,leaves at lower elevations had xeric morphological features such as increased epidermal thickness,sclerification and more developed metaxylem area.In conclusion,shifting of morphological features in below-and above-ground plant parts of C.serrulatus were linked to shifts in environmental factors along this elevation gradient,thus enabling the successful distribution of this species along this elevation gradient.展开更多
The biological function of the novel zinc-finger SWIM domain-containing protein family(ZSWIM)during embryonic development remains elusive.Here,we conducted a genome-wide analysis to explore the evolutionary processes ...The biological function of the novel zinc-finger SWIM domain-containing protein family(ZSWIM)during embryonic development remains elusive.Here,we conducted a genome-wide analysis to explore the evolutionary processes of the ZSWIM gene family members in mice,Xenopus tropicalis,zebrafish,and humans.We identified nine putative ZSWIM genes in the human and mouse genome,eight in the Xenopus genome,and five in the zebrafish genome.Based on multiple sequence alignment,three members,ZSWIM5,ZSWIM6,and ZSWIM8,demonstrated the highest homology across all four species.Using available RNA sequencing(RNAseq)data,ZSWIM genes were found to be widely expressed across different tissues,with distinct tissuespecific properties.To identify the functions of the ZSWIM protein family during embryogenesis,we examined temporal and spatial expression patterns of zswim family genes in Xenopus embryos.Quantitative real-time polymerase chain reaction(qRT-PCR)revealed that each member had a distinct expression profile.Whole-mount in situ hybridization showed that both zswim1 and zswim3 were maternally expressed genes;zswim5 and zswim6were expressed throughout embryogenesis and displayed dynamic expression in the brain,eyes,somite,and bronchial arch at the late tailbud stages;zswim7 was detected in the eye area;zswim8 showed a dynamic expression pattern during the tailbud stages,with expression detected in the brain,eyes,and somite;zswim9 was faintly expressed throughout embryonic development.This study provides a foundation for future research to delineate the functions of ZSWIM gene members.展开更多
Evidences show that the storage period greatly affects the quality of yogurt.In this study,three types of yogurt:control yogurt(CY),non-hydrolyzed potato powder yogurt(PPY)and enzymatically hydrolyzed potato powder yo...Evidences show that the storage period greatly affects the quality of yogurt.In this study,three types of yogurt:control yogurt(CY),non-hydrolyzed potato powder yogurt(PPY)and enzymatically hydrolyzed potato powder yogurt(EHPPY)were prepared at 42℃ for 5 h and stored for 28 days at 4℃.The yogurts were evaluated for quality characteristics at different storage periods.Negligible differences in pH values,titratable acidities and viable counts were detected in all three types of yogurt during storage.However,compared to other yogurts,EHPPY exhibited desirable water holding capacity,throughout the storage period.Apart from this,sensory properties and antioxidant activities(2-diphenyl-1-picryl-hydrazyl(DPPH)free radical scavenging activity and ferric reducing antioxidant power(FRAP))of EHPPY were also significantly improved during the storage period.Furthermore,the storage(G’)and loss(G”)modulus of PPY,EHPPY were lower than CY at 4℃ while a hysteresis loop was shown by all yogurts at the temperature range of 4-50℃ indicating higher G’(elasticity)than G”(viscosity).Based on our findings,EHPP could be an important functional ingredient in improving the quality and storage stability of yogurt for its production at an industrial level.展开更多
The beet armyworm(BAW),Spodoptera exigua(Lepidoptera:Noctuidae)is a highly destructive pest of vegetables and field crops.Management of beet armyworm primarily relies on synthetic pesticides,which is threatening the b...The beet armyworm(BAW),Spodoptera exigua(Lepidoptera:Noctuidae)is a highly destructive pest of vegetables and field crops.Management of beet armyworm primarily relies on synthetic pesticides,which is threatening the beneficial community and environment.Most importantly,the BAW developed resistance to synthetic pesticides with making it difficult to manage.Therefore,alternative and environment-friendly pest management tactics are urgently required.The use of pesticidal plant extracts provides an effective way for a sustainable pest management program.To evaluate the use of pesticidal plant extracts against BAW,we selected six plant species(Lantana camara,Aloe vera,Azadirachta indica,Cymbopogon citratus,Nicotiana tabacum,and Ocimum basilicum)for initial screening experiment.Four out of six plant species such as A.indica,N.tabacum,C.citratus and O.basilicum showed promising mortality of more than 50%.Therefore,we selected these four plant extracts for the subsequent experiments.Through contact bioassay,A.indica showed high mortality 66.63%,followed by the N.tabacum 53.33%,at 10%w/v concentration.Similarly,N.tabacum showed the highest mortality rate,66%at 10%w/v concentration,followed by the A.indica 46%through feeding bioassay.Furthermore,the feeding deterrence assay showed that C.citratus had a high antifeedant index(−50)followed by A.indica(−39),and N.tabacum(−28).In living plant assay,the N.tabacum extract showed a low mean damage score 3.6 on living cotton plant followed by C.citratus 4.5 and A.indica 5.5.Hence,extracts of three plant species provided promising results against the BAW,which can minimize the use of synthetic chemicals,particularly for small landholding farmers.Further studies are also required to evaluate the effects of these plant extract against BAW on cotton plants under field conditions to optimize the further use.展开更多
The study aimed to evaluate the ability of some terrestrial and aquatic plants for wastewater purification.Aquatic plants can remove pollutants from wastewater by consuming and accumulating various contaminants in diff...The study aimed to evaluate the ability of some terrestrial and aquatic plants for wastewater purification.Aquatic plants can remove pollutants from wastewater by consuming and accumulating various contaminants in different parts of plants.Different aquatic and terrestrial plants(Rosa sinensis,Typha latifolia,Ocimm bacilicum,Azolla pinnata,and Salvinia molesta)which have the ability to decrease water pollution were utilized in this study.The capability offive different species of plants was investigated by measuring chemical oxygen demand(COD),biological oxygen demand(BOD),electrical conductivity(EC),total dissolved solids(TDS),and pH of the medium.In this research,some aquatic and terrestrial plants were transplanted in wastewater plastic pots containing domestic wastewater with different ratios of 50%and 100%.Then,after 30 days,the physiological and biochemical parameters of plants were calculated to observe the effect of wastewater on plants.Results revealed higher chlorophyll and carotenoids in typha plants treated with 100%wastewater.The highest percentage of elimination in BOD(65%),COD(27%),TDS(72%),EC(83%),and pH(6.8%)was noted with the use of typha and azolla.Intriguingly,total soluble sugars,total free amino acids,and total proteins were found maximum in the hibiscus plant as compared to the other plants under 100%and 50%domestic wastewater treatment,while typha and ocimum showed lower values of these parameters irrespective of wastewater treatments.Moreover,the COD,BOD,TDS,EC,and pH trend was higher in 100%wastewater as compared to 50%wastewater.Taking into account the accumulation capacity of the tested plants especially typha can be efficiently used for the treatment of domestic wastewater.展开更多

文摘Background Pink bollworm,Pectinophora gossypiella(Saunders)(Lepidoptera:Gelechiidae)has become a poten-tial pest of cotton by causing substantial yield losses around the world including Pakistan.Keeping in view the facts like limited research investigations,unavailability,and high cost of artificial diet’s constituents and their premixes,the present research investigations on the dietary aspect of P.gossypiella were conducted.The larvae of P.gossypiella were reared on different diets that were prepared using indigenous elements.The standard/laboratory diet com-prised of wheat germ meal 34.5 g,casein 30.0 g,agar–agar 20.0 g,sucrose 10.0 g,brewer’s yeast 5.0 g,α-cellulose 1.0 g,potassium-sorbate1.5 g,niplagin 0.5 g,decavitamin 0.01 g,choline-chloride 0.06 g,maize-oil 3.30 g,honey 2.0 g,and water 730.0 mL.Alternatives to cotton bolls and wheat germ meal were okra seed sprouts,okra fruit,cottonseed meal,and okra seed meals,which were included in the study to introduce an efficient and economic mass-rearing system.Results The larval development completed in 19.68d±0.05 d with a weight of 20.18mg±0.20 mg at the fourth instar fed on the cottonseed meal-based diet instead of wheat germ meal based diet.On the same diet,84.00%±4.00%,17.24 mg±0.03 mg,and 7.76d±0.06 d were recorded as pupae formation,pupal weight,and pupal duration,respectively.Adult emergence,76.00%±1.00%was recorded from pupae collected from larvae raised on cottonseed meal-based diet.These male and female moths lived for 40.25d±0.10 d,and 44.34d±0.11 d,respectively.Females deposited 21.28±0.04 eggs per day with the viability of 65.78%±0.14%.The larval mortal-ity at the fourth instar was 37.20%±1.36%and malformed pupation of 12.00%±1.41%was recorded.Replacement of wheat germ meal with that of local meals(cottonseed and okra seed)in the standard laboratory diet has saved 463.80 to 467.10 PKR with 1.62 to 1.63 cost economic returns,respectively.Conclusion This research is of novel nature as it provides a concise and workable system for the economic and suc-cessful rearing of P.gossypiella under laboratory conditions.
文摘Background SRO(Similar to RCD1)genes family is largely recognized for their importance in the growth,develop-ment,and in responding to environmental stresses.However,genome-wide identification and functional characteri-zation of SRO genes from cotton species have not been reported so far.Results A total of 36 SRO genes were identified from four cotton species.Phylogenetic analysis divided these genes into three groups with distinct structure.Syntenic and chromosomal distribution analysis indicated uneven distribu-tion of GaSRO,GrSRO,GhSRO,and GbSRO genes on A2,D5 genomes,Gh-At,Gh-Dt,Gb-At,and Gb-Dt subgenomes,respectively.Gene duplication analysis revealed the presence of six duplicated gene pairs among GhSRO genes.In promoter analysis,several elements responsive to the growth,development and hormones were found in GhSRO genes,implying gene induction during cotton growth and development.Several miRNAs responsive to plant growth and abiotic stress were predicted to target 12 GhSRO genes.Organ-specific expression profiling demonstrated the roles of GhSRO genes in one or more tissues.In addition,specific expression pattern of some GhSRO genes dur-ing ovule development depicted their involvement in these developmental processes.Conclusion The data presented in this report laid a foundation for understanding the classification and functions of SRO genes in cotton.

文摘Background Cotton is a strategically important fibre crop for global textile industry.It profoundly impacts several countries’industrial and agricultural sectors.Sustainable cotton production is continuously threatened by the unpre-dictable changes in climate,specifically high temperatures.Breeding heat-tolerant,high-yielding cotton cultivars with wide adaptability to be grown in the regions with rising temperatures is one of the primary objectives of modern cotton breeding programmes.Therefore,the main objective of the current study is to figure out the effective breed-ing approach to imparting heat tolerance as well as the judicious utilization of commercially significant and stress-tolerant attributes in cotton breeding.Initially,the two most notable heat-susceptible(FH-115 and NIAB Kiran)and tolerant(IUB-13 and GH-Mubarak)cotton cultivars were spotted to develop filial and backcross populations to accom-plish the preceding study objectives.The heat tolerant cultivars were screened on the basis of various morphological(seed cotton yield per plant,ginning turnout percentage),physiological(pollen viability,cell membrane thermostabil-ity)and biochemical(peroxidase activity,proline content,hydrogen peroxide content)parameters.Results The results clearly exhibited that heat stress consequently had a detrimental impact on every studied plant trait,as revealed by the ability of crossing and their backcross populations to tolerate high temperatures.However,when considering overall yield,biochemical,and physiological traits,the IUB-13×FH-115 cross went over particularly well at both normal and high temperature conditions.Moreover,overall seed cotton yield per plant exhibited a posi-tive correlation with both pollen viability and antioxidant levels(POD activity and proline content).Conclusions Selection from segregation population and criteria involving pollen viability and antioxidant levels concluded to be an effective strategy for the screening of heat-tolerant cotton germplasms.Therefore,understanding acquired from this study can assist breeders identifying traits that should be prioritized in order to develop climate resilient cotton cultivars.
文摘The study focused on elaborating the role of GINS1 expression and its regulatory mechanisms in colon adenocarcinoma (COAD). Using the UALCAN informational index, GINS1 expression assessment unveiled a critical up- regulation in malignant cells that stood out from normal controls, suggesting its contribution to COAD expansion. Further dismantling GINS1 expression across various boundaries revealed unsurprising up-regulation in different malignant development stages, racial groups, genders, and age classes in COAD patients, characteristics for its imperative role in cancer progression. Moreover, this study investigated the promoter methylation status of GINS1, uncovering a critical uniqueness between COAD samples and normal controls. Analyzing promoter methylation across various clinical boundaries uncovered powerful variations, with particular methylation patterns seen across cancer stages, race groups, genders, and age groups. Survival analysis using the Kaplan-Meier (KM) plotter tool showed a colossal connection between GINS1 expression levels and overall survival (OS) in COAD patients, with low GINS1 expression interfacing with higher OS. Additionally, mutational examination using the cBioPortal stage revealed that no critical change was found in COAD. Overall, these findings revealed the complex contribution of GINS1 in COAD pathogenesis, underlining its actual limit as a prognostic biomarker and supportive therapeutic agent in COAD management.
文摘Continuously changing climate and availability of different rice genotypes make it necessary to find optimum time of sowing as well as suitable variety for cultivation to get maximum productivity under a specific set of climatic conditions. A field study was carried out to search out the suitable rice transplanting time for four different coarse genotypes under the semi-arid environment of Faisalabad. The experiment was conducted at Agronomic Research Area, University of Agriculture, Faisalabad and was laid out in randomized complete block design (RCBD) with split plot arrangement keeping transplanting time in main plots while rice genotypes in subplots. Variability among treatments was measured by Fisher’s ANOVA (P ≤ 5%) and LSD test was applied to compare the differences among treatments’ means. The ANOVA indicated statistically significant differences among genotypes as well as transplanting dates irrespective of all studied traits while interactive effects of both were found to be non-significant. NIBGE-1 performed best with maximum paddy yield of 6.05 t/ha while KSK-434 performed poor with paddy yield of 2.78 t/ha. Increased paddy yield and yield related parameters of all rice genotypes were recorded where transplantation was done on 25th of June. Generally, paddy yield decreased with delaying the transplanting time. The results suggested that NIBGE-1 can perform better under the semi-arid conditions of Faisalabad and last week of June might be the optimum time for nursery transplantation. It can also be further elucidated that late transplanting causes yield reduction which could not be recommended among farmers.
文摘Drinking water plays a major role regarding quality of human life because polluted water is the main cause of many types of diseases. Besides other pollutants, high concentration of uranium above a certain level in drinking water is also hazardous. As water and food are the main sources of uranium intake, it is absolutely imperative that their level of concentrations is monitored regularly for safety of the general public. In this regard, trace amount of uranium in drinking water samples collected from different locations of Faisalabadcity has been carried out using Neutron Induced Fission Track Technique. The water samples along with the standard of known uranium concentration were poured and dried over Lexan track detectors and then irradiated with thermal neutrons in Reactor. After etching, the tracks produced in the detectors as a result of 235U (n, f) reaction were counted under an optical microscope. The uranium concentration was determined by counting and comparing the observed fission track density in the samples and the standard. The observed uranium concentration in the studied water samples varied from (1.04 ± 0.30) μg·L-1 to (21.08 ± 2. 95) μg·L-1 with an average value of (7.39 ± 1.30) μg·L-1 and is found to be within safe limits as far as uranium related health hazards are concerned.

文摘Background High temperature stress at peak flowering stage of cotton is a major hindrance for crop potential.This study aimed to increase genetic divergence regarding heat tolerance in newly developed cultivars and hybrids.Fifty cotton genotypes and 40 F1(hybrids)were tested under field conditions following the treatments,viz.,high temperature stress and control at peak flowering stage in August and October under April and June sowing,respectively.Results The mean squares revealed significant differences among genotypes,treatments,genotype×treatment for relative cell injury,chlorophyll contents,canopy temperature,boll retention and seed cotton yield per plant.The genetic diversity among 50 genotypes was analyzed through cluster analysis and heat susceptibility index(HSI).The heat tolerant genotypes including FH-Noor,NIAB-545,FH-466,FH-Lalazar,FH-458,NIAB-878,IR-NIBGE-8,Weal-AGShahkar,and heat sensitive,i.e.,CIM-602,Silky-3,FH-326,SLH-12 and FH-442 were hybridized in line×tester fashion to produce F1 populations.The breeding materials’populations(40 F1)revealed higher specific combining ability variances along with dominance variances,decided the non-additive type gene action for all the traits.The best general combining ability effects for most of the traits were displayed by the lines,i.e.,FH-Lalazar,NIAB-878 along with testers FH-326 and Silky-3.Specific combining ability effects and better-parent heterosis were showed by the crosses,viz.,FH-Lalazar×Silky-3,FH-Lalazar×FH-326,NIAB-878×Silky-3,and NIAB-878×FH-326 for seed cotton yield and yield contributing traits under high temperature stress.Conclusion Heterosis breeding should be carried out in the presence of non-additive type gene action for all the studied traits.The best combiner parents with better-parent heterosis may be used in crossing program to develop high yielding cultivars,and hybrids for high temperature stress tolerance.
文摘Background Under K deficiency the uptake and distribution pattern in plant cells is mediated through different transport proteins and channels which were controlled by specific gene family.Therefore,a hydroponic experiment was conducted under control condition for testing the gene expression pattern of the K transporter under adequate and low K supply levels.After that,a 2-year field experiment was conducted to evaluate five selected cotton cultivars(four K-efficient cultivars,viz.,CIM-554,CYTO-124,FH-142,IUB-2013,and one K non-efficient,BH-212) screened from the initial hydroponics culture experiment and two levels of potassium(0 K_(2)O kg·ha^(-1) and 50 K_(2)O kg·ha^(-1)) were tested under reduced irrigation(50% available water content;50 AWC) and normal irrigation conditions(100% available water content;100 AWC).Result Results revealed that the transcript levels of GhHAK5aD in roots were significantly higher in K^(+) efficient cultivars than that in K^(+) non-efficient cultivars.The GhHAK5aD expression upon K^(+) deficiency was higher in roots but lower in shoots,indicating that GhHAK5aD could have a role in K^(+) uptake in roots,instead of transport of K^(+) from root to shoot.Similarly,under field conditions the cultivar FH-142 showed an increase of 22.3%,4.9%,2.4%,and 1.4% as compared with BH-212,IUB-2013,CYTO-124,and CIM-554,respectively,in seed cotton yield(SCY) with K application under reduced irrigation conditions.With applied K,the FH-142 showed an increase in net photosynthetic rate by 57.3% as compared with the rest of the cultivars under reduced irrigation over K control.However,the overall performance indicators of K-efficient cultivars like FH-142,CYTO-124,CIM-554,and IUB-2013 were better than BH-212(K in-efficient) under reduced irrigation conditions with applied K at 50 kg·ha^(-1).Fiber quality trait improved significantly with K application under water deficit.The increase in micronaire was 3.6%,4.7%,7.8%,3.4%,and 6.7% in BH-212,IUB-2013,CIM-554,CYTO-124,and FH-142,respectively,with K application at 50 kg·ha^(-1) over without K application under reduced irrigation conditions during the cotton growing season.Similarly,the cultivars FH-142 increased by 12% with K application under reduced irrigation as compared with other cultivars.The performance of K-efficient cultivars under reduced irrigation conditions was 30% better in SCY and quality traits with the application of K at 50 kg·ha^(-1) as compared with K-non-efficient cultivars.Similarly,water use efficiency(WUE)(40.1%) and potassium use efficiency(KUE)(20.2%) were also noted higher in case of FH-142 as compared with other cultivar with K application under reduced conditions.Conclusion Higher expression of GhHAK5aD gene was observed in K-efficient cultivars as compared with K-nonefficient cultivars in roots indicates that GhHAK5aD may be contributing to genotypic differences for K^(+) efficiency in cotton.K-efficient cotton cultivars can be used for the low-K environments and can also be recommended for general cultivars.

文摘In this research study, we have synthesized the bio-capped ZnO/g-C_(3)N_(4) nanocomposites by employing lemon juice(Citrus limon) as a stabilizer and mediator. Fruitfully, lemon juice which contains various acidic functional groups and citric acid has the capability to block the surface of g-C_(3)N_(4) from chemical reactivity and activated the surface of g-C_(3)N_(4) for various reactions. Consequently, the agglomeration behavior and controlled shape of g-C_(3)N_(4) has also been achieved. Our experimental results i.e. XRD,TEM, HRTEM, PL, FS, XPS, and PEC have confirmed that the lemon juice mediated and green g-C_(3)N_(4)(L-CN) have good performances and remarkable visible light photocatalytic activities as compared to the chemically synthesized g-C_(3)N_(4)(CN). Furthermore, the small surface area and low charge separation of g-C_(3)N_(4) is upgraded by coupling with Zn O nanoparticles. It is proved that the coupling of Zn O worked as a facilitator and photoelectron modulator to enhance the charge separation of g-C_(3)N_(4). Compared to pristine lemon-mediated green g-C_(3)N_(4)(L-CN), the most active sample 5Zn O/L-CN showed ~ 5-fold improvement in activities for ciprofloxacin(CIP) and methylene blue(MB) degradation. More specifically,the mineralization process and degradation pathways, and the mineralization process of ciprofloxacin(CIP) and methylene blue(MB) are suggested. Finally, our present novel research work will provide new access to synthesize the eco-friendly and bio-caped green g-C_(3)N_(4)nanomaterials and their employment for pollutants degradation and environmental purification.
文摘Crop production is greatly influenced by water and nutrition stress in semi-arid areas. Nitrogen and water are two most important yield limiting factors which effect economic production of wheat. Drought initiates when large area of land suffers from absence of precipitation for temporary periods. Though, not only water scarceness but also low relative humidity and high temperature are other climate factors which induce drought condition. Whereas Effective nutrition (nitrogen) maintains crops metabolic activities and has the potential to lessen drought stress. According to study carried out to explore the influence of varying levels of potential soil moisture deficit (PSMD) and nitrogen, an experiment was planned at agronomy research area, University of Agriculture, Faisalabad. It was laid out in randomized complete block design (RCBD) with split plot arrangement, and replicated thrice. Two factors: potential soil moisture deficit levels (I1: irrigation at 50 mm PSMD, I2: irrigation at 75 mm PSMD and I3: irrigation at 100 mm PSMD) were randomized in main plots, while sub plot nitrogen levels {control (No nitrogen), 50, 100 and 150 kg N ha-1} were randomized. Adequate nitrogen application along with sufficient irrigation management gave highest yield. Under water deficit conditions adequate nitrogen application ameliorates the drought stress. Highest plant height (86.27 cm), number of productive tillers (320.0), grain·spike-1(49.73), test weight (50.55 g) and grain yield (6.72 t·ha-1) were observed with 75 mm PSMD and 150 kg·ha-1 application of nitrogen. While radiation use efficiency for dry matter accumulation of wheat ranged from 1.53 - 1.87 g·MJ-1 whereas, radiation use efficiency for grain yield ranged from 0.421 - 0.473 g·MJ-1. Results revealed that highest RUE obtained I3 (75 mm PSMD) and nitrogen application at 150 kg·ha-1. Judicial application of irrigation and fertilizer not only boosts the yield but also saves resource and increases former output.
文摘Background Boron(B)deficiency is an important factor for poor seed cotton yield and fiber quality.However,it is often missing in the plant nutrition program,particularly in developing countries.The current study investigated B’s effect on growth,yield,and fiber quality of Bt(CIM-663)and non-Bt(Cyto-124)cotton cultivars.The experimental plan consisted of twelve treatments:Control(CK);B at 1 mg·kg^(−1) soil application(SB1);2 mg·kg^(−1) B(SB2);3 mg·kg^(−1) B(SB3);0.2%B foliar spray(FB1);0.4%B foliar spray(FB2);1 mg·kg^(−1) B+0.2%B foliar spray(SB1+FB1);1 mg·kg^(−1) B+0.4%B foliar spray(SB1+FB2);2 mg·kg^(−1) B+0.2%B foliar spray(SB2+FB1);2 mg·kg^(−1) B+0.4%B foliar spray(SB2+FB2);3 mg·kg^(−1) B+0.2%B foliar spray(SB3+FB1);3 mg·kg^(−1) B+0.4%B foliar spray(SB3+FB2).Each treat-ment has three replications,one pot having two plants per replication.Results B nutrition at all levels and methods of application significantly(P≤0.05)affected the growth,physiological,yield,and fiber quality characteristics of both cotton cultivars.However,SB2 either alone or in combination with foliar spray showed superiority over others,particularly in the non-Bt cultivar which responded better to B nutrition.Maxi-mum improvement in monopodial branches(345%),sympodial branches(143%),chlorophyll-a(177%),chlorophyll-b(194%),photosynthesis(169%),and ginning out turn(579%)in the non-Bt cultivar was found with SB2 compared with CK.In Bt cultivar,although no consistent trend was found but integrated use of SB3 with foliar spray performed relatively better for improving cotton growth compared with other treatments.Fiber quality characteristics in both cultivars were improved markedly but variably with different B treatments.Conclusion B nutrition with SB2 either alone or in combination with foliar spray was found optimum for improving cotton’s growth and yield characteristics.

文摘Salinity is one of the major abiotic factors that limit the growth and productivity of plants.Foliar application of plant growth regulators(PGRs)may help plants ameliorate the negative impacts of salinity.Thus,a field experiment was conducted at the Botanical Garden University of Balochistan,Quetta,to explore the potential role of PGRs,i.e.,moringa leaf extract(MLE;10%),proline(PRO;1μM),salicylic acid(SA;250μM),and thiourea(TU;10 mM)in ameliorating the impacts of salinity(120 mM)on Plantago ovata,an important medicinal plant.Salinity hampered plant photosynthetic pigments and metabolites but elevated oxidative parameters.However,foliar application of PGRs enhanced photosynthetic pigments,including Chl b(21.11%),carotenoids(57.87%)except Chl a,activated the defense mechanisms by restoring and enhancing the metabolites,i.e.,soluble sugars(49.68%),soluble phenolics(33.34%),and proline(31.47%),significantly under salinity stress.Furthermore,foliar supplementation of PGRs under salt stress led to a decrease of about 43.02%and 43.27%in hydrogen peroxide and malondialdehyde content,respectively.Thus,PGRs can be recommended for improved photosynthetic efficiency and metabolite content that can help to get better yield under salt stress,with the best and most effective treatments being those of PRO and MLE to predominately ameliorate the harsh impacts of salinity.

文摘We developed a molecular marker for MAS of mungbean resistant varieties against CLS from the consensus sequence(MB-CLsRG)of identified RGAs(MB-ClsRCaG1 and MB-ClsRCaG2).The MB-CLsRG sequence-specific primer pair was used to screen Cercospora leaf spot(CLS)resistant varieties of mungbean in genomic analysis that showed congruency with phenotypic screening.Validation of molecular marker linkage with CLS resistance was performed using rtPCR in transcriptomic analysis.The sequenced PCR products showed 100%homology with MB-CLsRG sequence and putative disease resistance proteins that confirmed the linkage of molecular marker with CLS resistance in mungbean.The antifungal potential of MB-CLsRG gene encoding protein was assessed.The MB-CLsRG gene sequence was cloned in the E.coli expression vector for recombinant protein production.The recombinant protein was then investigated for its in vitro antifungal potential against Cercospora canescens.The in vitro investigation showed strong antifungal activity of recombinant protein as it restricted the growth of fungal mycelial mass.The results validated the linkage of developed marker with CLS-resistant mungbean varieties;therefore,it can be used to screen resistant varieties from a large population in MAS.Moreover,the recombinant protein of the MB-CLsRG gene sequence revealed antifungal potential,which proved the gene sequence could be suitable to use in transgenic plants technology to develop fungal-resistant transgenic crops.
文摘Calotropis procera(Aiton)W.T.Aiton,belonging to the family Apocynaceae,is C3 evergreen plant species in arid and semi-arid areas of the Punjab Province,Pakistan.It grows in a variety of habitats like salt affected and waterlogged area,desert/semi-desert,roadside,wasteland,graveyard,forest,crop field,coastline,and river/canal bank.A total of 12 populations growing in different ecological regions were sampled to evaluate their growth,physio-biochemical,and anatomical responses to specific environmental condition.Population adapted to desert/semi-desert showed vigorous growth(plant height,shoot length,and number of leaves),enhanced photosynthetic level(chlorophyll a,chlorophyll b,carotenoids,and total chlorophyll),and apparent anatomical modifications such as increased stem radius,cuticle thickness,storage parenchyma tissues(cortex and pith),and vascular bundles in stems,while the maximum of midrib and lamina thickness,epidermal cells,cuticle thickness,cortical proportion,abaxial stomatal density,and its area in leaves.There was high plasticity in structural and functional features of these populations,which enable them to survive and tolerate under such hot and dry desert environment.Population of saline areas exhibited very critical modifications to sustain under salt prone environment.At physiological level,it possesses the maximum amount of organic osmolytes(glycine betaine and proline)and antioxidants(superoxide dismutase(SOD),catalase(CAT),and peroxidase(POD)),while at anatomical level,it showed intensive sclerification,large phloem region(inner and outer),pith parenchyma cells,and metaxylem vessels in stems and leaves.The population of dry mountains showed very distinctive features,such as increased shoot ionic contents(K+and Ca2+),collenchyma and sclerenchyma thickness in stems,trichomes size,and numerous small stomata on abaxial surface of leaves.It is concluded that no definite or precise single character can be taken as a yardstick for adjudging the biomass production in this rubber bush weed population.
文摘The imperative aspect of the CRISPR/Cas9 system is a short stretch of 20 nucleotides of gRNA that control the overall specificity.Due to the small size,the chance of its multiple occurrences in the genome increases;however,a few mismatches are tolerated by the Cas9 endonuclease activity.An accurate and careful in silico-based off-target prediction while target selection is preferred to address the issue.These predictions are based on a comprehensive set of selectable parameters.Therefore,we investigated the possible off-target prediction and their screening in StERF3 gene-edited potato plants while developing StERF3-loss-of-function mutants using CRISPR/Cas9 approach.The 201 off-targets for the selected targets of the StERF3 gene were predicted,and 79 werefiltered as potential off-targets.Of these 79,twenty-five off-targets showed scores with defined cut-off values<0.5 and were analyzed in Sterf3-edited potato plants compared to wild-type plants.No off-targeting was found to have occurred in edited plants.
文摘Chrysopogon serrulatus(false beard-grass)is a dominant component of vegetation in the foothills of the Himalayas.To study whole plant morphology,individuals of C.serrulatus were collected from three plots at each of six locations spanning from 400 to 1,400 m.The population colonizing the highest elevation modifications in different plant organs.Roots showed increased metaxylem number and area.In the stem,especially outside of the vascular tissue,there was intensive sclerification indicative of increased xeromorphy as a survival strategy.At the highest elevation,leaves were wider;aerenchyma formation and increased sclerification were noted in the leaf sheath;and a greater proportion of storage parenchyma was observed in the leaf blade,all indicators of succulence.In contrast,leaves at lower elevations had xeric morphological features such as increased epidermal thickness,sclerification and more developed metaxylem area.In conclusion,shifting of morphological features in below-and above-ground plant parts of C.serrulatus were linked to shifts in environmental factors along this elevation gradient,thus enabling the successful distribution of this species along this elevation gradient.

文摘The biological function of the novel zinc-finger SWIM domain-containing protein family(ZSWIM)during embryonic development remains elusive.Here,we conducted a genome-wide analysis to explore the evolutionary processes of the ZSWIM gene family members in mice,Xenopus tropicalis,zebrafish,and humans.We identified nine putative ZSWIM genes in the human and mouse genome,eight in the Xenopus genome,and five in the zebrafish genome.Based on multiple sequence alignment,three members,ZSWIM5,ZSWIM6,and ZSWIM8,demonstrated the highest homology across all four species.Using available RNA sequencing(RNAseq)data,ZSWIM genes were found to be widely expressed across different tissues,with distinct tissuespecific properties.To identify the functions of the ZSWIM protein family during embryogenesis,we examined temporal and spatial expression patterns of zswim family genes in Xenopus embryos.Quantitative real-time polymerase chain reaction(qRT-PCR)revealed that each member had a distinct expression profile.Whole-mount in situ hybridization showed that both zswim1 and zswim3 were maternally expressed genes;zswim5 and zswim6were expressed throughout embryogenesis and displayed dynamic expression in the brain,eyes,somite,and bronchial arch at the late tailbud stages;zswim7 was detected in the eye area;zswim8 showed a dynamic expression pattern during the tailbud stages,with expression detected in the brain,eyes,and somite;zswim9 was faintly expressed throughout embryonic development.This study provides a foundation for future research to delineate the functions of ZSWIM gene members.

文摘Evidences show that the storage period greatly affects the quality of yogurt.In this study,three types of yogurt:control yogurt(CY),non-hydrolyzed potato powder yogurt(PPY)and enzymatically hydrolyzed potato powder yogurt(EHPPY)were prepared at 42℃ for 5 h and stored for 28 days at 4℃.The yogurts were evaluated for quality characteristics at different storage periods.Negligible differences in pH values,titratable acidities and viable counts were detected in all three types of yogurt during storage.However,compared to other yogurts,EHPPY exhibited desirable water holding capacity,throughout the storage period.Apart from this,sensory properties and antioxidant activities(2-diphenyl-1-picryl-hydrazyl(DPPH)free radical scavenging activity and ferric reducing antioxidant power(FRAP))of EHPPY were also significantly improved during the storage period.Furthermore,the storage(G’)and loss(G”)modulus of PPY,EHPPY were lower than CY at 4℃ while a hysteresis loop was shown by all yogurts at the temperature range of 4-50℃ indicating higher G’(elasticity)than G”(viscosity).Based on our findings,EHPP could be an important functional ingredient in improving the quality and storage stability of yogurt for its production at an industrial level.
文摘The beet armyworm(BAW),Spodoptera exigua(Lepidoptera:Noctuidae)is a highly destructive pest of vegetables and field crops.Management of beet armyworm primarily relies on synthetic pesticides,which is threatening the beneficial community and environment.Most importantly,the BAW developed resistance to synthetic pesticides with making it difficult to manage.Therefore,alternative and environment-friendly pest management tactics are urgently required.The use of pesticidal plant extracts provides an effective way for a sustainable pest management program.To evaluate the use of pesticidal plant extracts against BAW,we selected six plant species(Lantana camara,Aloe vera,Azadirachta indica,Cymbopogon citratus,Nicotiana tabacum,and Ocimum basilicum)for initial screening experiment.Four out of six plant species such as A.indica,N.tabacum,C.citratus and O.basilicum showed promising mortality of more than 50%.Therefore,we selected these four plant extracts for the subsequent experiments.Through contact bioassay,A.indica showed high mortality 66.63%,followed by the N.tabacum 53.33%,at 10%w/v concentration.Similarly,N.tabacum showed the highest mortality rate,66%at 10%w/v concentration,followed by the A.indica 46%through feeding bioassay.Furthermore,the feeding deterrence assay showed that C.citratus had a high antifeedant index(−50)followed by A.indica(−39),and N.tabacum(−28).In living plant assay,the N.tabacum extract showed a low mean damage score 3.6 on living cotton plant followed by C.citratus 4.5 and A.indica 5.5.Hence,extracts of three plant species provided promising results against the BAW,which can minimize the use of synthetic chemicals,particularly for small landholding farmers.Further studies are also required to evaluate the effects of these plant extract against BAW on cotton plants under field conditions to optimize the further use.
文摘The study aimed to evaluate the ability of some terrestrial and aquatic plants for wastewater purification.Aquatic plants can remove pollutants from wastewater by consuming and accumulating various contaminants in different parts of plants.Different aquatic and terrestrial plants(Rosa sinensis,Typha latifolia,Ocimm bacilicum,Azolla pinnata,and Salvinia molesta)which have the ability to decrease water pollution were utilized in this study.The capability offive different species of plants was investigated by measuring chemical oxygen demand(COD),biological oxygen demand(BOD),electrical conductivity(EC),total dissolved solids(TDS),and pH of the medium.In this research,some aquatic and terrestrial plants were transplanted in wastewater plastic pots containing domestic wastewater with different ratios of 50%and 100%.Then,after 30 days,the physiological and biochemical parameters of plants were calculated to observe the effect of wastewater on plants.Results revealed higher chlorophyll and carotenoids in typha plants treated with 100%wastewater.The highest percentage of elimination in BOD(65%),COD(27%),TDS(72%),EC(83%),and pH(6.8%)was noted with the use of typha and azolla.Intriguingly,total soluble sugars,total free amino acids,and total proteins were found maximum in the hibiscus plant as compared to the other plants under 100%and 50%domestic wastewater treatment,while typha and ocimum showed lower values of these parameters irrespective of wastewater treatments.Moreover,the COD,BOD,TDS,EC,and pH trend was higher in 100%wastewater as compared to 50%wastewater.Taking into account the accumulation capacity of the tested plants especially typha can be efficiently used for the treatment of domestic wastewater.
