AIM: To evaluate the in vitro immunomodulation capacity of various non-pathogenic yeast strains and to investigate the ability of some of these food grade yeasts to prevent experimental colitis in mice.METHODS: In vitro immunomodulation was assessed by measuring cytokines [interleukin (IL)-12p70,IL-10,tumor necrosis factor and interferon γ] released by human peripheral blood mononuclear cells after 24 h stimulation with 6 live yeast strains (Saccharomyces ssp.) and with bacterial reference strains.A murine model of acute 2-4-6-trinitrobenzene sulfonic acid (TNBS)-colitis was next used to evaluate the distinct prophylactic protective capacities of three yeast strains compared with the performance of prednisolone treatment.RESULTS: The six yeast strains all showed similar non-discriminating anti-inflammatory potential when tested on immunocompetent cells in vitro .However,although they exhibited similar colonization patterns in vivo ,some yeast strains showed significant anti-inflammatory activities in the TNBS-induced colitis model,whereas others had weaker or no preventive effect at all,as evidenced by colitis markers (body-weight loss,macroscopic and histological scores,myeloperoxidase activities and blood inflammatory markers).CONCLUSION: A careful selection of strains is required among the biodiversity of yeasts for specific clinical studies,including applications in inflammatory bowel disease and other therapeutic uses. 展开更多

Somatic molecular changes and histo-pathological features of colorectal cancer in Tunisia

AIM:To determine correlations between family history,clinical features and mutational status of genes involved in the progression of colorectal cancer(CRC).METHODS:Histo-pathological features and molecular changes[KRAS,BRAF and CTNNB1 genes mutations,microsatellite instability(MSI)phenotype,expression of mismatch repair(MMR)and mucin(MUC)5AC proteins,mutation and expression analysis of TP53,MLH1promoter hypermethylation analysis]were examined in a series of 51 unselected Tunisian CRC patients,10 of them had a proven or probable hereditary disease,on the track of new tumoral markers for CRC susceptibility in Tunisian patients.RESULTS:As expected,MSI and MMR expression loss were associated to the presence of familial CRC(75%vs 9%,P<0.001).However,no significant associations have been detected between personal or familial cancer history and KRAS(codons 12 and 13)or TP53(exons 4-9)alterations.A significant inverse relationship has been observed between the presence of MSI and TP53 accumulation(10.0%vs 48.8%,P=0.0335)in CRC tumors,suggesting different molecular pathways to CRC that in turn may reflect different environmental exposures.Interestingly,MUC5AC expression was significantly associated to the presence of MSI(46.7%vs 8.3%,P=0.0039),MMR expression loss(46.7%vs8.3%,P=0.0039)and the presence of familial CRC(63%vs 23%,P=0.039).CONCLUSION:These findings suggest that MUC5AC expression analysis may be useful in the screening of Tunisian patients with high risk of CRC. 展开更多

Polycrystalline of[(Na_(0.535)K_(0.480))_(0.966)Li_(0.058)](Nb_(0.90)Ta_(0.10))O_(3)samples were prepared using the high-temperature solid-state reaction technique.X-ray diffraction(XRD)analysis indicates the formation of a single-phase with orthorhombic structure.AC impedance plots were used as tool to analyze the electrical behavior of the sample as a function of frequency at different temperatures.The AC impedance studies revealed the presence of grain effect,from 425℃onwards.Complex impedance analysis indicated non-Debye type dielectric relaxation.The Nyquist plot showed the negative temperature coefficient of resistance(NTCR)characteristic of NKLNT.The AC conductivity results were used to correlate with the barrier hopping(CBH)model to evaluate the binding energy(W_(m)),the minimum hopping distance(R_(min)),the density of states at Fermi level(N(E_(f))),and the activation energy of the compound. 展开更多
