To explore mechanism and protective effect of rosiglitazone on myocardial ischemia reperfusion(I/R) injury.Methods:A total of 48 male Japanese white big-ear rabbits were randomly divided into control group(A),I/R group(B),low dose of rosiglitazone group(C),high dose of rosiglitazone group(D).Plasma concentration of and also reduced the concentration of plasma serum creatine kinase(CK),CK-MB.high-sensitivity C-reactive protein(hsCRP).ultrasuperoxide dismutase(SOD),malondialdehyde(MD.A).lactic acid glutathione skin peroxidase (C-SH-PX).nitric oxide(NO)and endothelin(ET) were measured 1 h later after I/R.Twenty-four hours after I/R the hearts were harvested for pathological and ultrastructural analysis.Area of myocardial infarction were tested.Results:Plasma concentration of CK,Ck-MB.hsCRP,NO. MDA and ET were decreased in C,D group compared with group B.Plasma concentration of T-SOD and GSH-Px were increased significantly in C.D group compared with group B.Compared with group B.pathological and ullraslructural changes in C and D group were slightly.There was significant difference in myocardial infarction area between group C.D and group B(P【0.05). Myocardial infarction area and arrhythmia rate were lower in group C,D compare with group B. Rosiglitazone may protect myocardium from I/R injury by enhancing T-SOD and GSH-Px concentration,inhibit inflammatory reaction,and improve endothelial function. 展开更多

Background Several studies have assessed the association between cerebral palsy(CP)and assisted reproductive technology(ART),but the results remain controversial.We conducted a meta-analysis to evaluate the risk of CP after ART compared with natural conceptions and to examine CP risk separately in ART singletons,multiples and preterm births.Methods Web-based databases(PubMed,Embase,the Cochrane Library,and Web of Science)were searched until November 22,2020.Studies which compare CP rates after ART with natural conceptions were included.The Newcastle–Ottawa Scale was used to assess the quality of the included studies.Effect estimates were extracted and combined using the fixed-effects or random-effects model depending on the heterogeneity test.Results There were nine studies included in the meta-analysis.The included studies were of moderate or high quality.A significantly higher risk of CP[odds ratio(OR)=2.17,95%confidence interval(CI)1.72–2.74]was found in ART children(n=89,214)compared with naturally conceived children(n=4,160,745).The significantly higher risk decreased when data were restricted to singletons(OR=1.36,95%CI 1.16–1.59)and disappeared when data were restricted to multiples(OR=1.05,95%CI 0.86–1.29)or preterm births(OR=1.53,95%CI 0.66–3.56).Subgroup and sensitivity analyses indicated that the overall results were robust.Conclusions The risk of CP is increased more than two-fold after ART.This increased risk is largely due to increased rates of multiple birth and preterm delivery in ART children. 展开更多

In this study,we have designed a more clinically powerful anti-caries treatment by applying the amelogenin-derived peptide QP5 to the antibacterial carrier material chitosan in a hydrogel(CS-QP5 hydrogel),and characterized its effects on the inhibition of a cariogenic biofilm and the promotion of the remineralization of the initial caries lesions.The results indicated that the CS-QP5 hydrogel sustainably inhibited the growth of the Streptococcus mutans biofilm,lactic acid production and the metabolic activity over a prolonged period of time.Moreover,the CS-QP5 hydrogel promoted the remineralization of early enamel lesions,which were indicated by surface microhardness(,polarized light microscopy and transverse microradiography.In conclusion,the CS-QP5 hydrogel shows good potential for caries control in the clinic because of its antibacterial effects as well as the remineralization of initial enamel carious lesions even in a biofilm model over a prolonged period of time. 展开更多

Background:Echinococcosis is a parasitic zoonosis caused by Echinococcus larvae parasitism causing high mortality.The Tibetan Region of Sichuan Province is a high prevalence area for echinococcosis in China.Understanding the geographic distribution pattern is necessary for precise control and prevention.In this study,a spatial analysis was conducted to explore the town-level epidemiology of echinococcosis in the Sichuan Tibetan Region and to provide guidance for formulating regional prevention and control strategies.Methods:The study was based on reported echinococcosis cases by the end of 2017,and each case was geo-coded at the town level.Spatial empirical Bayes smoothing and global spatial autocorrelation were used to detect the spatial distribution pattern.Spatial scan statistics were applied to examine local clusters.Results:The spatial distribution of echinococcosis in the Sichuan Tibetan Region was mapped at the town level in terms of the crude prevalence rate,excess hazard and spatial smoothed prevalence rate.The spatial distribution of echinococcosis was non-random and clustered with the significant global spatial autocorrelation(I=0.7301,P=0.001).Additionally,five significant spatial clusters were detected through the spatial scan statistic.Conclusions:There was evidence for the existence of significant echinococcosis clusters in the Tibetan Region of Sichuan Province,China.The results of this study may assist local health departments with developing better prevention strategies and prompt more efficient public health interventions. 展开更多

Concentrated growth factor regulates the macrophage-mediated immune response

Concentrated growth factor(CGF)is a promising regenerative material that serves as a scaffold and adjunct growth factor for tissue engineering.The host immune response,particularly macrophage activity,plays a critical role in injury repair and tissue regeneration.However,the biological effect of CGF on the immune response is not clear.To enrich the theoretical groundwork for clinical application,the present study examined the immunoregulatory role of CGF in macrophage functional activities in vitro.The CGF scaffold appeared as a dense fibrin network with multiple embedded leukocytes and platelets,and it was biocompatible with macrophages.Concentrated bioactive factors in the CGF extract enhanced THP-1 monocyte recruitment and promoted the maturation of suspended monocytes into adherent macrophages.CGF extract also promoted THP-1 macrophage polarization toward the M2 phenotype with upregulated CD163 expression,as detected by cell morphology and surface marker expression.A cytokine antibody array showed that CGF extract exerted a regulatory effect on macrophage functional activities by reducing secretion of the inflammatory factor interleukin-1b while inducing expression of the chemokine regulated on activation,normal T cell expressed and secreted.Mechanistically,the AKT signaling pathway was activated,and an AKT inhibitor partially suppressed the immunomodulatory effect of CGF.Our findings reveal that CGF induces a favorable immune response mediated by macrophages,which represents a promising strategy for functional tissue regeneration. 展开更多
